Cryopreservation of sperm in marine fish ArchiMer

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Most extenders used in marine fish are saline or sugar solutions. From the investigated cryoprotectants, dimethyl sulphoxide (DMSO) generally leads to the best results....

Induction of ovulation and stimulation of spermation by LHRH or an LHRH analog associated or not with pimozide. Effects on some teleost fish. ArchiMer

Effectiveness of LHRH and one of its analogs (LHRH-A = D-Ala super(6) ethylamide) on induction of ovulation and stimulation of spermiation is reviewed here. LHRH was also combined with pimozide, an anti-dopaminergic agent, to try to cancel dopamine inhibition of gonatotropin (GtH) release. LHRH-A alone (50 mu g/kg) did not initiate ovulation but only oocyte maturation. When pimozide (10 mg/kg) and LHRH-A (50 mu g/kg) were combined, gonadotropic hormone secretion was significantly increased, causing ovulation and stimulating spermiation. This combination advanced ovulation in trout. Trout egg quality was markedly altered by the pimozide treatment; it remains to be tested in carp. Marine fish spermatozoa: racing ephemeral swimmers ArchiMer

After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. The cost of such 'hyperactivity' is a very rapid consumption of intracellular ATP that outstrips the supply....
